While Suika might have seemed like nothing more than a run-of-the-mill comic relief character when she was initially introduced, her nearsightedness and the issues that it caused for her instantly made her quite endearing to fans of the series.
So, when Senku gave her contact lenses so that she could finally see clearly for the first time in her life, Suika's emotional reaction was a thing to behold and easily established itself as one of the most heartwarming moments in the series. Initially serving under the ranks of Tsukasa and tasked to find and report on Senku, Gen Asagiri gets wooed in the smartest manner by Senku when he finally showcases the capabilities of the Kingdom of Science, with a bottle of cola serving as proof of why scientific achievements should remain paramount in the Stone World as opposed to brute force and outdated mentalities.
After coming to his senses, Gen decides to switch sides and becomes a valuable ally for Senku, serving as a tactical and strategic member of the Five Wise Generals who always had stellar points to bring up at any given moment... especially during a war that was critical when it came to deciding the path that the new world would tread.
4 Tsukasa Shishio

The primary antagonist of Dr. Stone who ends up being a thorn in Senku's side with his outdated ideals and misguided views on how the Stone World should evolve, Tsukasa is one of the most powerful and intimidating characters in the entire series. However, while his kingdom might've been strong from a physical perspective, the lack of scientific prowess meant that his time at the top was destined to be brief from the very start.
Thankfully, he finally comes to his senses and decides to partner up with his rival once he ends up losing the higher ground during the Stone Wars. The ensuing quest to rescue his daughter along with Senku ends up eliminating their rivalry once and for all, with Tsukasa finally seeing the light and joining the Kingdom of Science as a valuable ally.
3 Yuzuriha Ogawa

Forming the original trio of friends who ended up turning into stone during the Petrification Event in 2019, Yuzuriha is a rather unique — if not somewhat cliched — character who also serves as the love interest of Taiju in the series. After realizing the threat that Tsukasa posed to the new world, Senku decided upon a plan that would ensure the safety of both their friends by telling them to join Tsukasa's Kingdom as spies.
After Tsukasa's defeat, Yuzuriha ended up joining the Kingdom of Science and performed the duties of a seamstress, weaving clothes for the people of this kingdom.
2 Taiju Oki

His personality is so unlike what one would expect from the lead of a shonen series, and yet his character doesn't suffer as a result of this — if anything, it ends up elevating his character to new heights and turns him into one of the most interesting characters in the series.
Watching this mad genius reach a large number of scientific breakthroughs and put them to good use in practical situations becomes the highlight of the series, making it infinitely clear why he's the biggest draw of Dr. Stone by a country mile.
