Chapter 205, the final chapter of the manga, is set far into the future. It focuses on the descendants of the series' characters throughout the story as we know it. Aoba Hashibira, the great-grandson of Inosuke Hashibira and Aoi Kanzaki, is a working botanist who is conducting research on the Blue Spider Lily. In his work, he confirms that the flower blooms during the daytime exclusively, only two or three days a year. However, due to a mistake, all the flowers withered.

Chapter 127 tells the origin of Muzan becoming a demon. When he was young, Muzan was afflicted with a deadly disease, one that would kill him before he turned 20 years old. Out of anger due to his worsening condition, Muzan murdered his doctor before completing his course of treatment. Muzan then discovered that while the medicine did strengthen his body, the side effects made sunlight fatal to him, and he developed a thirst for human flesh. This is how Muzan came to be a demon.

Muzan remembered the experimental prototype medicine that the doctor was using on him and the components that comprised it, but he had no idea of how to create this "Blue Spider Lily" medicine. Consequently, he never learned what was necessary to complete his course of treatment for his disease. He scoured the land in search of the Blue Spider Lilies, but to no avail. He hoped that if he found them, he could figure out how to complete his course of treatment.

Muzan's Recipe For Immortality

Muzan Kibutsuji Side Profile Demon Slayer

Muzan's ultimate goal is to attain an immortal body. He wanted to be able to walk in the sunlight and live forever. In order to gain this, he needed two things: the Blue Spider Lily and the body of a demon who could withstand the sun.

In Chapter 127, Lady Tamayo writes a letter to Tanjiro, theorizing that Nezuko may be able to walk in the sunlight soon. Since Nezuko has "conquered the sun", this piques the interest of Muzan in his quest for immortality. After all, she would be the second ingredient he needs to achieve his long-sought-after goal.

However, since the Blue Spider Lilly was found to only bloom during the day, that may explain why Muzan never found them after spending so long looking. As a demon, Muzan would obvioulsy not be able to go out and find them due to the devastating effects of sunlight on his body. So ultimately, the cruel villain's quest would never be fulfilled.
